5-Amino-1MQ (5-Amino-1-methylquinolinium) is a cell-permeable, selective, and potent inhibitor of the enzyme Nicotinamide N-methyltransferase (NNMT). NNMT is a cytosolic enzyme predominantly expressed in adipose tissue and the liver, where it plays a critical role in energy metabolism.
By inhibiting NNMT, 5-Amino-1MQ increases intracellular levels of Nicotinamide Adenine Dinucleotide (NAD+), effectively shifting the metabolic state of the cell toward increased energy expenditure and enhanced fat oxidation.
Key Research Applications & Benefits
-
NNMT Inhibition: Specifically targets the NNMT enzyme to prevent the depletion of NAD+ and S-adenosylmethionine (SAM), crucial for cellular energy.
-
Metabolic Rate Augmentation: Research suggests a significant increase in the basal metabolic rate (BMR) of white adipose tissue, promoting the "browning" of fat.
-
Muscle Hypertrophy & Repair: Studies indicate that 5-Amino-1MQ may stimulate the proliferation of satellite cells, aiding in the regeneration and growth of skeletal muscle.
-
Anti-Obesity Potential: Extensively used in models of diet-induced obesity to study the reduction of body weight and white adipose tissue without reducing food intake.
-
Senolytic Properties: Emerging data explores its role in combating age-related metabolic decline and improving insulin sensitivity in aging subjects.
